  • Despite the success of anti-retroviral therapy (ART), the persistence of HIV reservoirs is a major barrier to overcome before finding a lasting cure.
  • The HIV-1 transactivation protein (Tat) binds the HIV mRNA transactivation responsive element (TAR), regulating transcription and reactivation from latency.
  • While N-803 alone did not reactivate virus production, its administration after CD8+ lymphocyte depletion induced the most robust and persistent.

    Introduction

Although the Berlin, London, and Düsseldorf adult patients appeared to have experienced a sterilizing human immunodeficiency virus (HIV) cure1, the treatment regimen involved toxic chemotherapy followed by hematopoietic stem cell transplantation, which is not feasible for most patients due to the high risk and expense. In HIV-infected adult patients, proviral reservoirs are extremely stable during long-term antiretroviral therapy (ART), and become the driving force for viral rebound after analytic treatment interruption (ATI)2,3,4,5,6. Although the timing of viral genome integration is uncertain in acute HIV infection, adult non-human primate models of simian immunodeficiency virus (SIV) suggest that proviral reservoirs are established as early as 3 days post infection7,8. Mother-to-child HIV transmission can occur during in utero at late pregnancy, at the time of delivery or in the days or weeks after birth through breastfeeding9,10. Current interventions to prevent in utero HIV infection have been highly successful, yet ~150,000 children are still postnatally infected with HIV every year11. Although early initiation of ART in infants may red
